While it's definitely easier to work with pixel sizes at the start, I occasionally got some feedback on a design that suggests the fonts in general just need to be "a little bigger" or "a little smaller". While I save some time up front, by not having to calculate relative em sizes, after a request like that, I certainly then have to spend a good amount of time touching every single pixel-sized font and line-height to accommodate even such a "little" request.
